mRNA expressions of heat shock protein 90 α, β in peripheral blood mononuclear cells of patients with polymyositis or dermatomyositis

摘要:

目的 探讨多发性肌炎/皮肌炎(PM/DM)患者外周血单个核细胞(PBMC)中热休克蛋白90(HSP90)α,βmRNA表达与PM/DM患者对糖皮质激素(GC)疗效的相关性。方法 采用逆转录-聚合酶链反应(RT-PCR)方法检测29例GC治疗敏感、15例GC治疗抵抗的PM/DM患者及25例正常对照PBMC中HSP90 α,βmRNA的表达。结果 GC治疗敏感和抵抗的PM/DM患者PBMC中HSP90 α,βmRNA的表达均显著高于正常对照,且GC治疗抵抗的PM/DM患者PBMC中HSP90,βmRNA的表达显著高于GC治疗敏感的PM/DM患者。结论 HSP90α,βmRNA 表达水平升高可能与PM/DM患者的GC治疗抵抗相关。

Abstract:

Objective To explore the relationship between expressions of heat shock protein90(HSP90) α,βmRNA in peripheral blood mononuclear cell (PBMC) of patients with polymyositis/dermatomyositis(PM/DM) and the efficacy of glucocorticoid(GC) therapy.Methods Reverse transcription-polymerase chain reaction(RT-PCR) was used to semiquantite the expressions of HSP90) α,βin PBMC of 29 cases of PM/DM patients sensitive to GC,15 cases of PM/DM patients resistant to GC and 25 cases of the normal control. Results The expressions of α,βmRNA in PBMC both of PM/DM patients sensitive to GC and resistant to GC were significantly higher than that of the normal control, and the expressions of HSP90α,βmRNA of PM/DM patients resistant to GC was significantly higher than that of PM/DM patients sensitive to GC. Conclusion The higher expression of HSP90α,βmRNA in PBMC of PM/DM patients may be correlated with PM/DM resistant to GC therapy.
