糠秕马拉色菌对角质形成细胞增殖的作用

摘要:

[摘要] 目的:检测不同浓度糠秕马拉色菌对角质形成细胞增殖的影响及其可能机制。方法:用MTT法检测不同浓度糠秕马拉色菌 (糠秕马拉色菌:HaCaT细胞数量比分别为0:1、1:1、5:1、10:1、20:1、30:1、40:1、60:1、80:1、100:1)作用下角质形成细胞株HaCaT细胞增殖的变化,同时用MTT法观察HIF-1α的特异性抑制剂YC-1(10μM)、VEGF抑制剂CBO-P11(2μM)、VEGFR-2的中和性抗体MAB3571(5μg/ml)是否可以拮抗低浓度糠秕马拉色菌(20:1)对HaCaT细胞的促增殖作用;蛋白免疫印迹检测低浓度糠秕马拉色菌(20:1)和YC-1(10μM)作用下HaCaT细胞HIF-1α、VEGF、VEGFR-2的蛋白质表达。结果:低浓度糠秕马拉色菌促进HaCaT细胞增殖,而高浓度糠秕马拉色菌诱导HaCaT细胞坏死及凋亡; 低浓度糠秕马拉色菌(20:1)对HaCaT细胞的促增殖作用可分别被YC-1(10μM)、 CBO-P11(2μM)、MAB3571(5μg/ml)所拮抗;低浓度糠秕马拉色菌(20:1)促进HaCaT细胞表达HIF-1α、VEGF、VEGFR-2蛋白质表达,但是此作用可以被HIF-1α的特异性抑制剂YC-1(10μM)阻断。结论:低浓度糠秕马拉色菌可以通过激活HIF-1α/VEGF/VEGFR-2信号通路促进角质形成细胞增殖。

Abstract:

[Abstract]: Objective to determine the effect and possible mechanism of different concentration of malassezia furfur on the proliferation of keratinocytes. Methods MTT assay was used to determine the effect of different concentration of malassezia furfur(the ratio of malassezia furfur and HaCaT cells as 0:1;1:1;5:1;10:1;20:1;30:1;40:1;60:1;80:1;100:1) on the proliferation of HaCaT cells. Migration assay was also used to determine the antagonism effect of 10μM YC-1(pecific inhibitor of HIF-1α), 2μM CBO-P11(pecific inhibitor of VEGF), 5μg/ml MAB3571(neutralizing antibody of VEGFR-2)on the proliferation of HaCaT cells promoted by malassezia furfur(20:1). Westen blotting was used to determine the protein expression of HIF-1α, VEGF, VEGFR-2 . Results Low concentration of malassezia furfur promoted the proliferation of HaCaT cells, while high concentration of malassezia furfur promoted the apoptosis and necrosis of HaCaT cells. The promoted proliferation of HaCaT cells by malassezia furfur (20:1) was inhibited by YC-1(10μM), CBO-P11(2μM)or MAB3571(5μg/ml). Malassezia furfur (20:1) induced the protein expression of HIF-1α, VEGF, VEGFR-2, which was antagonized by YC-1(10μM). Conclusions Low concentration of malassezia furfur can promote the proliferation of HaCaT cells through the activation of HIF-1α/VEGF/VEGFR-2 pathway.
