液基微量稀释法体外测定脓肿分枝杆菌的药物敏感性

In vitro testing of antimicrobial susceptibility of Mycobacterium abscessus with broth mirodilution method

摘要:

目的 测定3株脓肿分枝杆菌(2个临床株和1个标准株)对抗菌药物的敏感性。方法 根据美国临床和实验室标准协会M24-A肉汤微量稀释法测定脓肿分枝杆菌对阿米卡星、多西环素、克拉霉素、磺胺甲基异噁唑、左氧氟沙星、氯法齐明、氨苯砜7种抗菌药物的敏感性。结果 3株脓肿分枝杆菌的MIC范围为:阿米卡星均为32μg/ml;多西环素均大于32μg/ml;克拉霉素为1~8μg/ml;磺胺甲基异噁唑均大于128μg/ml;左氧氟沙星为16~32μg/ml;氯法齐明为1~8μg/ml;氨苯砜均大于128μg/ml。各菌株对阿米卡星、多西环素、磺胺甲基异噁唑、左氧氟沙星、氨苯砜表现为中间耐药或耐药。3株中仅有1株对克拉霉素和氯法齐明敏感。结论 美国临床和实验室标准协会M24-A微量稀释法具有操作简便,实用性强,准确性高的优点。不同脓肿分枝杆菌对7种常用抗菌药物的敏感性存在差异。

Abstract:

Objective To evaluate the susceptibility of three strains of M. abscessus to antimicrobial agents. Methods Clinical Laboratory Standard Institute Broth microdilution method M24-A was applied to test the susceptibility of M. abscessus strains to amikacin, doxycycline, clarithromycin, sulfamethoxine, levofloxacin, clofazimine and dapsone. Results MIC ranges of three strains of M. abscessus were: amikacin 32μg/ml, doxycycline more than 32μg/ml, clarithromycin 1~8μg/ml, sulfamethoxine more than 128μg/ml, levofloxacin 16~32μg/ml, clofazimine 1~8μg/ml, dapsone more than 128μg/ml. Of the three strains, only one was sensitive to clarithromycin and clofazimine. Conclusion CLSI M24-A microdilution method is reliable, practical and easy to use in the clinical laboratory. The sensitivity of various strains of M. abscessus to the above mentioned antimicrobial agents is different.
