β1,4-半乳糖基转移酶-Ⅰ在瘢痕疙瘩中的表达

Expression of β-1,4-galactosyltransferase-Ⅰin keloid tissue

摘要: 目的探讨 β1,4-半乳糖基转移酶-Ⅰ(beta-1,4-galactosyltransferase-Ⅰ, β-1,4-GalT-Ⅰ)在瘢痕疙瘩发病机制中的作用.方法取瘢痕疙瘩组织标本10份,同时以正常人皮肤6份、增生性瘢痕7份做对照,测定组织中羟脯氨酸含量,通过逆转录-聚合酶链反应(RT-PCR)和图像定量分析,检测组织中 β-1,4-GalT-Ⅰ的表达水平.结果瘢痕疙瘩组织中羟脯氨酸含量明显高于正常人皮肤组织(P<0.01). β-1,4-GalT-ⅠmRNA指数值在瘢痕疙瘩、正常人皮肤及增生性瘢痕组织分别为(0.2995±0.0825),(0.0451±0.0287),(0.2769±0.0975),瘢痕疙瘩组织与正常人皮肤比较,差异有统计学意义(P<0.01),与增生性瘢痕比较差异无统计学意义(P>0.05).瘢痕疙瘩与增生性瘢痕组织中 β-1,4-GalT-Ⅰ表达的高低与其中胶原的含量均呈正相关(r=0.9496,P<0.01;r=0.8202,P<0.05).结论 β-1,4-GalT-Ⅰ在瘢痕疙瘩纤维化进程中可能起着调节作用.

Abstract: Objective To investigate the role of β-1,4-galactosyltransferase-Ⅰ( β-1,4-GalT-Ⅰ) in the pathogenesis of keloid.Methods Tissue samples were resected from 10 patients with keloid,6 normal human controls and 7 patients with hypertrophic scar.The content of hydroxyproline was determined by alkali hydrolyzation method,and the expression of β-1,4-GalT-Ⅰgene by reverse transcription-PCR and image analysis technique.Results The content of hydroxyproline and mRNA expression of β-1,4-GalT-Ⅰwere significantly higher in the keloid tissues than in the normal controls(10.98±1.55μg/mg vs 5.38±0.47μg/mg, 0.2995±0.0825 vs 0.0451±0.0287,respectively,both P<0.01).However,no significant difference was found in either of hydroxyproline content or β-1,4-GalT-ⅠmRNA expression between tissue samples of keloid and hypertrophic scar(both P>0.05).There was a positive correlation between the mRNA expression of β-1,4-GalT-Ⅰand hydroxyproline content in the tissue of keloid(r=0.9496,P<0.01)and of hypertrophic scar.Conclusion β-1,4-GalT-Ⅰmay play a modulatory role in the fibrotic process of keloid tissue.
