基质细胞衍生因子-1及受体CXCR4在阴道念珠菌病小鼠模型中的表达

Expression of stroma-derived factor 1 and its receptor CXCR4 in a murine model of vaginal candidiasis

摘要: 目的 检测基质细胞衍生因子-1(SDF-1)及其受体CXCR4在阴道念珠菌病小鼠模型阴道组织及腰引流淋巴结中的表达情况。方法 采用雌激素化阴道念珠菌病小鼠模型,雌性昆明小鼠(80只)随机分四组,A组(雌激素处理感染组)、B组(雌激素处理未感染组)、C组(未用雌激素处理感染组)、正常对照组(5只)。前三组又再分为接种白念珠菌后第2、4、7、14和21天组(各5只)。接种后不同时间点随机取小鼠阴道组织及腰淋巴结,以RT-PCR及免疫组化SABC法检测SDF-1及CXCR4 mRNA及蛋白水平的表达。结果 (1)RT-PCR:与正常小鼠相比,雌激素(B组)对SDF-1及CXCR4 mRNA表达无影响。①小鼠阴道组织:与B组相比,A组小鼠接种后各时间点均见SDF-1 mRNA表达明显增加(F = 114.87,P < 0.01),第14天CXCR4 mRNA明显增加(t = 15.61,P < 0.01);与正常小鼠相比,C组小鼠接种后第2,4, 7天,SDF-1 mRNA明显增加;第4 ~ 21天CXCR4 mRNA均明显增加。②腰淋巴结:与B组相比,A组小鼠接种后第2天和7 ~ 21天SDF-1 mRNA明显增加(F = 865.62,P < 0.01),CXCR4 mRNA于各时间点表达均增加(F = 161.30,P < 0.01);与正常小鼠相比,C组小鼠接种后SDF-1 mRNA表达无明显改变(P > 0.05);CXCR4 mRNA于第7天和14天明显增加(P < 0.05)。(2)免疫组化:A组小鼠阴道黏膜中SDF-1及CXCR4表达在接种后第2天即开始增加,第7天时达高峰,第14天和21天开始下降,但均明显高于对照组(P < 0.05)。结论 感染组小鼠阴道黏膜及腰淋巴结中SDF-1及CXCR4的mRNA及蛋白水平均明显升高。

Abstract: Objective To examine the expressions of stroma-derived factor 1 (SDF-1) and its receptor CXCR4 in murine vaginal tissue and lumbar lymph nodes during an experimental Candida albicans vaginal infection. Methods A murine model of vaginal candidiasis was established. Eighty female mice were randomly divided into 4 groups, 25 in group A (estrogen-treated mice inoculated with referrence C. albicans strain), 25 in group B (estrogen-treated mice inoculated with phosphate buffered solution), 25 in group C (untreated mice infected with C. albicans) and 5 untreated mice free of infection in the control group. The first 3 groups were equally divided into 5 subgroups, including 2-, 4-, 7-, 14- and 21-day groups. The mRNA and protein expression of SDF-1 and CXCR4 in the murine vaginal tissue and draining lumbar lymph nodes was assessed by RT-PCR and immunohistochemistry, respectively, 2, 4, 7, 14 and 21 days after the inoculation of C. albicans. Results No significant difference was observed in the mRNA expression of SDF-1 or CXCR4 between group B and the control group. Compared with group B, the mRNA level of SDF-1 in group A increased both in vaginal tissue from day 2 to 21 (F = 114.87, P < 0.01) and in lumbar lymph nodes on day 2, 7, 14 and 21; increased mRNA expression of CXCR4 was observed in vaginal tissue on day 14 (t = 15.61, P < 0.01) and lumbar lymph nodes from day 2 to 21 (F = 161.30, P < 0.01) in group A. In group C, the mRNA expression of SDF-1 significantly increased in vaginal tissue on day 2, 4 and 7 but remained unchanged in lumbar lymph tissues through the test period (P > 0.05), and an significant increase was seen in mRNA expression of CXCR4 in vaginal tissue from day 4 to 21 and in lumbar lymph tissues on day 7 and 14 in comparison with the control group (all P < 0.05). As immunohistochemistry showed, the expression levels of SDF-1 and CXCR4 in vaginal tissue of group A commenced to increase on day 2, reached a peak on day 7, then decreased on day 14 and 21 following the infeciton (P < 0.05), but they were consistently higher than those of the control group through the study period. Conclusion An increase in mRNA and protein expressions of SDF-1 and CXCR4 is noticed in vaginal tissue and lumbar lymph nodes of C. albicans-infected mice.
