T细胞因子4基因在毛囊毛乳头细胞中的表达

摘要: 目的 探讨T细胞因子4(TCF4)基因在毛囊毛乳头细胞中的表达。方法 对斑秃患者和正常人头皮组织进行原位杂交检测;对凝集性生长和非凝集性生长的人毛乳头细胞分别进行细胞免疫化学检测和用RT-PCR方法从上述两种细胞的总RNA扩增TCF4基因。结果 组织内mRNA原位杂交证实,TCF4基因在正常头皮组织毛囊毛乳头细胞表达,而在斑秃组织毛囊的毛乳头细胞不表达。细胞免疫化学和RT-PCR发现,TCF4基因在凝集性生长期毛乳头细胞中表达,而在非凝集性生长期毛乳头细胞不表达。结论 TCF4基因在生长期毛乳头细胞中表达,毛囊周期性生长与wnt信号有关。

Abstract: Objective To investigate the expression of T cell factor-4 (TCF4) gene in dermal papilla cells of hair follicles. Methods The expression of TCF4 gene was examined by in situ hybridization in scalp tissues of patients with alopecia areata and normal human controls. The protein and mRNA expressions of TCF4 were detected by immunochemistry and RT-PCR method, respectively, in aggregated and non-aggregated human dermal papilla cells. Results As shown by in situ hybridization, TCF4 gene was expressed in the dermal papilla cells from healthy controls, but not in those from patients with alopecia areata. Both cell immunochemistry and RT-PCR showed that TCF4 gene expressed in aggregated dermal papilla cells, but not in non-aggregated dermal papilla cells. Conclusions TCF4 gene is expressed in dermal papilla cells. The growth cycle of follicles may be related to wnt signal.
