表没食子儿茶素没食子酸酯对体外培养的人毛囊生长和毛乳头细胞增殖的影响

Effects of EGCG on the growth of human hair follicles and cell proliferation of human dermal papilla cells(DPCs) in vitro

摘要: 目的 研究表没食子儿茶素没食子酸酯(EGCG)对体外培养的人毛囊生长和毛乳头细胞增殖的影响。方法 显微分离人毛囊,在加入不同质量浓度EGCG (0 ~ 250 mg/L)的Williams E培养基中培养,观察毛囊生长情况;利用两步酶法分离培养人毛乳头细胞,加入不同质量浓度的EGCG (0 ~ 500 mg/L),48 h后,利用噻唑蓝(MTT)法检测细胞增殖,流式细胞仪检测细胞周期。结果 与空白对照组比较,0.1,1 mg/L的EGCG对体外培养的毛囊生长有明显促进作用(P < 0.05);MTT显示15.6,31.2, 62.5 mg/L的EGCG能够明显促进毛乳头细胞的增殖(P < 0.05);流式细胞仪检测7.8,15.6,31.2和62.5 mg/L的EGCG作用48 h后,S期和G2/M期细胞比率明显增加,G0/G1期细胞比率明显减少,细胞增殖指数明显增加(由5.25%增至5.64%,14.61%,32.51%,37.35%),细胞增殖指数与EGCG浓度呈显著正相关(r = 0.933)。结论 一定浓度的EGCG能够促进体外培养的人毛囊生长及人毛乳头细胞增殖。

Abstract: Objective To evaluate the effects of (-)-epigallocatechin-3-gallate (EGCG) on the growth of human hair follicles and proliferation of cultured human dermal papilla cells (DPCs) in vitro. Methods Isolated hair follicles were cultured in 0.5 ml Williams Medium E with EGCG at 10 different concentrations (0, 0.01, 0.1, 1, 10,15.6, 31.25, 62.5, 125, 250 mg/L). The growth of hair follicles was observed and recorded on day 1 and 10 during the culture. Human DPCs were isolated by two-step method from hair follicles, and cultured with various concentrations (0 - 500 mg/L) of EGCG. After 48 hours of culture, MTT assay was used to detect the proliferating rates of cultured DPCs, flow cytometry to analyze the cell cycle. Results Compard with the untreated controls, the growth of human hair follicles EGCG was stimulated at the concentrations of 0.1 and 1 mg/L (P < 0.05), and the proliferation of cultured human DPCs was accelerated by the treatment with EGCG at the concentrations from 15.6 to 62.5 mg/L(P < 0.05). An increase of cell proportion in S, G2 /M phases and decrease in G1 phase were observed in DPCs treated with EGCG at the concen- trations between 7.8 and 62.5 mg/L (P < 0.05). Proliferation index of DPCs was increased from 5.25% to 5.64%, 14.61%, 32.51%, 37.35% by EGCG of 7.8, 15.6, 31.2 and 62.5 mg/L, respectively; and the increasing effect was positively correlated with the concentration of EGCG (r = 0.933). Conclusion EGCG of given concentrations can accelerate the growth of human hair follicles and proliferation of cultured human DPCs.
