皮肤恶性黑素瘤中诱导型一氧化氮合酶mRNA及其蛋白的表达

The mRNA and protein expression of inducible nitric oxide synthase in cutaneous malignant melanoma

摘要： 目的 探讨皮肤恶性黑素瘤中诱导型一氧化氮合酶(iNOS)的表达及其临床意义.方法 免疫组化方法检测30例皮肤恶性黑素瘤患者肿瘤组织中iNOS蛋白的表达,逆转录聚合酶链反应(RTPCR)检测其中6例患者肿瘤组织和肿瘤邻近正常组织中iNOS mRNA的表达.结果 iNOS在肿瘤组织中无论蛋白或mRNA阳性表达率皆明显高于邻近正常组织(P<0.05).其中iNOS蛋白在无转移的恶性黑素瘤中阳性表达率为69.2%,有转移的恶性黑素瘤中阳性表达率为100%.结论 iNOS表达异常在恶性黑素瘤发生、发展与转移过程中可能起重要作用.

Abstract: Objective To investigate the mRNA and protein expression of inducible nitric oxide synthase(iNOS) and its clinical significance in cutaneous malignant melanoma(MM).Methods The iNOS protein expression was detected in tissue sections from 30 MM patients using immunohistochemistry method.Reverse transcription polymerase chain reaction(RT-PCR) was used to measure the iNOS mRNA expression in MM tissues and adjacent normal tissues from 6 of the 30 MM patients.Results The positive rates of iNOS protein and mRNA were both significantly higher in tumorous samples than those in normal tissues(86.7% vs 20.0%,P=0.000; 4/6 vs 1/6,P=0.024).The positive rate of iNOS protein was 69.2% in MM without metastasis,significant lower than that(100%) in MM with metastasis.Conclusion The abnormal expression of iNOS may play an important role in the occurrence,development and metastasis of MM.
