重组腺病毒介导金黄色葡萄球菌肠毒素A基因在小鼠B16黑素瘤细胞中的表达

The Expression of Staphylococcal Enterotoxin A(SEA) Gene on Murine B16 Melanoma Cells Mediated by Replication-defective Recombinant Adenovirus Vector

摘要: 目的 研究金黄色葡萄球菌肠毒素A(SEA)基因通过重组腺病毒介导转染入B16黑素瘤细胞中的表达情况。方法 用RT-PCR和流式细胞仪检测SEA基因在B16黑素瘤细胞中的表达。结果 在转染SEA基因的B16黑素瘤细胞中检测到SEA基因的转录,并于转染后24h开始有SEA蛋白的表达,到72h表达量达最高值87.22,之后减弱,直到168h仍有少许表达。结论 SEA基因可以在B16黑素瘤细胞中表达,为下一步黑素瘤的基因治疗奠定基础。

Abstract: Objective To investigate staphylococcal enterotoxin A (SE A) gene expression on murine B16 melanoma cells mediated by replication-defective recombinant adenovirus. Methods SEA expression on the B16 melanoma cells was detected by reverse transcription-polymerase chain reaction (RT-PCR) and flow cytometry (FCM). Results RT-PCR and FCM proved that SEA could specifically express on the B16 melanoma cells. Conclusion The successful expression of SEA gene on the B16 melanoma cells might be used for the targeted treatment of mali gnant melanoma.
