T淋巴细胞对小鼠接触变应性光皮炎的影响

摘要: 目的 探讨T淋巴细胞在接触变应性光皮炎(CAPD)发病中的作用.方法 选用BALB/c纯系雌性小鼠建立氯丙嗪所致的CAPD模型,采用腹腔注射抗小鼠CD4、CD8单克隆抗体(mAb)方法,激发后48h测小鼠耳肿度,局部皮损行HE染色,计单一核细胞浸润数.ELISA法测小鼠血清白介素10(IL-10)、干扰素γ(IFN-γ)水平.结果 腹腔注射抗CD4mAb组,IL-10水平与阴性对照组(仅光激发,不做光诱导)接近(P>0.05),而IFN-γ水平较阴性对照组高(P<0.05);与腹腔注射抗CD8mAb组相比,IL-10、IFN-γ水平均降低,但耳肿度及单一核细胞浸润数该两组接近,且该两组IL-10、IFN-γ水平均低于阳性对照组(P<0.05);联合注射抗CD4、CD8mAb组,IL-10、IFN-γ水平,耳肿度及单一核细胞浸润数均接近阴性对照组(P>0.05).结论 在氯丙嗪诱导的CAPD模型中,CD4+Th1和CD8+T淋巴细胞可通过上调IFN-γ水平正调节该反应;而CD4+Th2淋巴细胞可通过上调IL-10水平下调该反应.

Abstract: Objective To explore the role of T lymphocytes in contact allergic photodermatitis(CAPD). Methods BALB/c female mice were used to establish the model of CAPD induced by chlorpromazine. The mice were injected intraperitoneally with anti-mouse CD4?CD8 mAb. The ear swelling was evaluated at 48h after the challenge. The lymphocytes were counted in H.E section of the swellon ear. The levels of IFN-γ and IL-10 in serum were detected by ELISA. Results Comparing with the negative control, anti-CD4 mAb injected mice had almost the same level of IL-10 (P>0.05), but had a higher level of IFN-γ (P<0.05). The levels of IL-10 and IFN-γ were significantly lower in anti-CD4 mAb injected mice than those in anti-CD8 mAb injected mice (P<0.05). However there was no difference in the ear swelling and mononuclear cell counting between these two groups. Furthermore, the levels of IL-10 and IFN-γ were lower in these two groups than those in the positive control group. There was no difference in the levels of IL-10 and IFN-γ, ear swelling and mononuclear cell counting between combination injection (anti-CD4 plus anti-CD8) group and the negative control group. Conclusions In the model of CAPD induced by chlorpromazine, CD4+Th1 and CD8+T lymphocytes up-regulate the reaction by increasing the level of IFN-γ. However, CD4+Th2 lymphocytes down-regulate the reaction by increasing the level of IL-10.
