白念珠菌胞壁溴化十六烷基三甲铵甘露聚糖下调白介素6和8的研究

The Cell-Wall-Mannan of C.albicans Downregulate Both IL-6and IL-8 Expression in Human Peripheral Blood Mononuclear Cells:An In Vitro Study

摘要: 目的 探讨白念珠菌胞壁溴化十六烷基三甲铵(CTAB)甘露聚糖影响脂多糖诱导人外周血单一核细胞(PBMC)产生的IL-6、IL-8的效应。方法 3种浓度的CTAB甘露聚糖(1.00mg/mL、0.10mg/mL、0.01mg/mL)体外与人PBMC共孵育预刺激24h,然后加入脂多糖(50μg/mL)再孵育24h,收集上清液;在24h、48h时收集3种浓度CTAB甘露聚糖刺激组的上清液;阳性对照为脂多糖(50μg/mL),并设不加刺激剂的空白对照,酶联免疫法检测各组上清液中白介素-6、IL-8(IL-6、8)的含量。结果 在24h、48h3种浓度CTAB甘露聚糖刺激组均未检出IL-6、IL-8;阳性对照脂多糖组IL-6含量为(478.507±24.876)ng/mL,IL-8含量为(529.655±53.279)ng/mL;阴性对照未检出IL-6、IL-8;1.00mg/mLCTAB甘露聚糖+脂多糖组IL-6含量为(85.620±16.058)ng/mL,IL-8为(123.940±20.319)ng/mL;0.10mg/mLCTAB甘露聚糖+脂多糖组IL-6含量为(210.086±27.874)ng/mL,IL-8为(206.798±31.878)ng/mL;0.01mg/mLCTAB甘露聚糖+脂多糖组IL-6含量为(201.387±32.396)ng/mL,IL-8为(203.133±36.012)ng/mL.结论 白念珠菌胞壁CTAB甘露聚糖对脂多糖诱导人PBMC产生IL-6、IL-8具有下调作用。

Abstract: Objective To investigate the potential effect of cetyltrimethyl ammonium bromide(CTAB)separated mannan of cell wall from Candida albicans on the production of IL-6 and IL-8 in human peripheral blood mononuclear cells(PBMC)induced by lipopoly saccharide(LPS).Methods PBMCs were pretreated with different concentrations of CTAB mannan(1.000mg/mL、0.100mg/mL、0.010mg/mL)for 24h.LPS(50μg/mL)was added and co-incubated for 24h.And at 48h,the supernatants were collected.At 24h and 48h,only the super-natants of stimulated by CTABmannan were collected.LPS(50μg/mL)was the positive control,unstimulated culture medium the negative control.The contents of IL-6 and IL-8 in the supernatants were determined by ELISA.Results At 24h and 48h,no IL-6 and IL-8 were detected in 3 different concentration-CTAB mannan groups.LPS could induce IL-6(478.507±24.876ng/mL),IL-8(529.655±53.279ng/mL).The contents of IL-6 and IL-8 of negative control were not detectable.In 1.000mg/mL CTAB mannan+LPS group the contents of IL-6were(85.620±16.058ng/mL,P=0.004),IL-8 were(123.940±20.319ng/mL,P=0.011).In 0.100mg/mL CTAB mannan+LPS group,IL-6(210.086±27.874ng/mL,P=0.007),IL-8(206.798±31.878ng/mL,P=0.022).In 0.010mg/mL CTAB mannan+LPS group,IL-6(201.387±32.396ng/mL,P=0.014),IL-8(203.133±36.012ng/mL,P=0.015).Conclusion CTAB mannan of cell wall from Candida albicans could downregulate the production of IL-6 and IL-8 from human peripheral blood mononuclear cells induced by LPS.
