人中性粒细胞防御素对念珠菌的抗菌活性研究

In Vitro Antifungal Activity of Defensin from Human Neutrophils Against Some Candida spp.

摘要: 目的 探讨防御素在机体抗念珠菌中的作用及其抗菌机制。方法 用5%乙酸提取人中性粒细胞中的酸溶性成分,采用凝胶过滤层析获得人中性粒细胞防御素(human neutrophil peptide,HNP),检测HNP对白念珠菌M1012R、白念珠菌NIHA-207、乳酒念珠菌、热带念珠菌、季也蒙念珠菌等5株念珠菌的抗菌活性。同时用电镜观察防御素作用后念珠菌的超微结构变化。结果 采用Sephadex G100及Biogel P10凝胶过滤层析可获得纯化的HNP。HNP对白念珠菌M1012R、NIHA-207、乳酒念珠菌、热带念珠菌、季也蒙念珠菌等菌有抗菌活性,其活性随防御素浓度升高而增强。电镜观察见防御素作用后的念珠菌表面有明显的凹陷和穿孔,胞膜与胞壁分离及空泡形成等改变。结论 HNP对念珠菌有较强的抗菌活性。

Abstract: Objective To investigate the role and mechanism of defensin from human neutrophilic granulocytes against some Candida spp. Methods Human neutrophil peptide(HNP) were isolated and purified by using gel filtration chromatography. The antifungal activity of HNP against Candida albicans M1012R, NIH A-207, Candida tropicalis, Candida kefyr, and Candida guilliermondii was tested. The above fungi were observed under electron microscope after exposing to HNP. Results HNP was succesfully isolated and purified by Sephadex G100 and Bio-gel P10 gel filtration chromatography, and concentration-dependent candidcidal activity to Candida albicans M1012R, NIH A-207, Candida kefyr, Candida tropicalis, and Candida guilliermondii of HNP was demonstreated in vitro. After exposing to HNP the Candida spp. were examined by electron microscope, profound cell wall changes such as cell surface collapse and perforation were noted,the separation of cell membrane from cell wall and formation of vacuole were also observed. Conclusion Human neutrophil peptide exerted potent antifungal activity against some Candida spp.
