表皮生长因子对游离毛囊生长的影响

Effects of Epidermal Growth Factor on Human Hair Follicle Growth in vitro

摘要: 目的 通过毛囊器官培养观察表皮生长因子(EGF)对人头皮毛囊生长的影响。方法 用WilliamsE无血清培养基进行人头皮毛囊游离培养,分为2组,EGF组(43根)和对照组(43根)。结果 EGF20μg/L组在加药后1~3d毛干平均生长0.22mm/d,对照组平均生长0.16mm/d,两组差异有显著性(P<0.05),EGF对毛囊生长有更明显的促进作用。但EGF组毛囊于培养的第4~6天即出现退行期的形态改变并发生扭曲,毛干停止生长,对照组于培养第10天后才发生形态变化。组织学上,培养第10天EGF组毛囊下部外毛根鞘增厚,杵状结构形成,而对照组仍为生长期形态。3H-亮氨酸掺入实验显示EGF组毛囊上皮中的放射活性(370c/min)高于对照组(271.14c/min)(P<0.05),而毛干中的放射活性(173.19c/min)与对照组(158.49c/min)无明显差异。结论 EGF对毛囊的生长有促进作用,主要是促进毛囊上皮的生长,并且诱导毛囊提前由生长期进入退行期。

Abstract: Objective To investigate the effects of epidermal growth factor (EGF) on hair follicle growth. Methods Cultured human hair follicles were divided into two groups:EGF-treated group (EGF 20 ng/mL) and control group. The length and morphology of follicles were measured by inverted microscope daily. 3H-leucine incorporation and histologic studies were applied. Results The rates of hair shaft and hair follicle elongation significantly increased in the EGF-treated group during day 1 to day 3 in comparison with the control group. Histologically, the cells of the lower outer root sheath were thickened and club hair-like structure was formed. 3H-leucine incorporation showed that the radioactivity in hair follicle epithelium was higher in EGF-treated group (370 c/min)than that in control group(271.14 c/min) (P<0.05). No difference was found in terms of the radioactivity in hair shaft between EGF-treated group(173.19 c/min) and control group(158.49 c/min). Conclusion It is suggested that EGF might stimulate the growth of hair follicle epithelium, and induce anagen transforming into catagen prematurely.
