白介素2激活人外周血单一核细胞抗白念珠菌活性的研究

摘要: 目的 研究在体外人白介素1(IL-1)、重组人白介素2(IL-2)、重组人γ-干扰素 (IFN-γ)、重组人肿瘤坏死因子 (TNF)能否激活人外周血单一核细胞 (PBMC),使之具有抗白念珠菌活性.方法用Ficoll Hypaque密度梯度离心法,分离36例正常人PBMC.将PBMC分别同一定浓度的IL-1、IL-2、IFN-γ、TNF共同培养一定时间后,收集培养后的细胞与一定浓度的白念珠菌共同培养 2h,检测每毫升中白念珠菌菌落形成单位 (CFU/mL).结果 IL-2组的PBMC有抑制白念珠菌生长的活性,而IL-1、IFN、TNF均无刺激PBMC产生抗白念珠菌生长活性的作用;PBMC及IL-2本身均无抑制白念珠菌生长的活性;IL-2激活的PBMC的抗白念珠菌活性作用与IL-2的浓度及共同培养时间有关.结论 IL-2及IL-2激活的PBMC可能在宿主抵抗白念珠菌感染中起重要作用,同时为治疗念珠菌病提供了一个新的思路.

Abstract: Objective To investgate the ability of cytokine activated human peripheral blood mononuclear cell (PMBC) in inhibiting Candida albicans in vitro Methods PBMCs of thirty six volunteers were obtained by Ficoll Hypaque density centrifugation and cultured for 7 days in cell wells During the culture period different cytokines including,recombinant human tumor necrosis factor (TNF),interleukin-1(IL 1),gamma interferon(IFN-γ) and interleukin-2(IL-2) were added respectively Then,the treated PBMCs were incubated with C-albicans in vitro for 2h to determine the number of colony forming unit of C-albicans per milliliter(cfu/mL) Results Compared to that of controls,there was a significant decrease of C-albicans cfu/mL in IL-2-activated group (P<0.01) Whereas,no inhibitory effect was observed in IL 1,IFNγor TNFαstimulated groups The inhibitory effect of IL-2 activated PBMC was related to the concentration of IL-2 and the duration of culture PBMC was not able to inhibit C-albicans growth directly(P>0.05) IL-2 itself failed to inhibit the growth of C-albicans Conclusion These data demonstrate that IL-2 is capable of stimulating antifungal activity of PBMC which may play an important role in host defensing against C-albicans infections and provide a new approach for the treatment
