解脲支原体第一群的分型研究

Serotyping of Biovar One of Ureaplasma urealyticum by Molecular Biological Methods

摘要: 目的 利用分子生物学方法对解脲支原体(Ureaplasma urealyticum,Uu)第一群进行分型,建立简便、可靠的分型方法。方法 设计分型引物,用PCR和限制性内切酶消化的方法进行分型。结果 设计了解脲支原体的分型引物UMS51|UMAU1和UMS51|UMAU3,前者扩增出Uu1型和Uu6型,后者扩增出Uu3型和Uu14型。对于Uu1型和Uu6型,用限制性内切酶消化的方法区分。结论 根据MB抗原基因的N端,利用PCR结合限制性内切酶消化的方法可以对解脲支原体第一群进行分型,但不能将Uu3型与Uu14型区分开。

Abstract: Objective To set up serotyping methods of Ureaplasma urealyticum by molecular biological techniques.Methods Primers for typing were designed and different serotypes of Ureaplasma urealyticum were identified by PCR and restriction endonuclease digestion.Results PCR primers UMS51, UMAU1 and UMS51,UMAU3 was specific to Uu1,Uu6 and Uu3, Uu14 respecitively. To distinguish Uu1 and Uu6, restriction endonuclease digestion were used.Conclusion By PCR and restriction endonuclease digestion on the basis of DNA sequence of N terminal of multiple-banded antigen gene, serovars of biovar one of Uu can be distinguished but Uu3 and Uu14 can not be distinguished.
