糠秕孢子菌与培养人角朊细胞的相互作用

Interaction of Malassezia furfur to Cultured Human Keratinocytes

摘要: 为了探讨糠秕孢子菌与角朊细胞的相互作用,建立了菌体与人角朊细胞共同培养系统,将形态和机能研究紧密结合:①定时将培养物作扫描和透射电镜观察;②用Alamarblue荧光分析法观测条件培养液对角朊细胞生长的影响;③用ELISA法定量检测条件培养液中IL1α和bFGF.结果发现:①随时间经过角朊细胞将菌体粘附、吞噬,有明显的形态变化特点;②条件培养液可促进角朊细胞的生长,从中检出高水平的IL1α和bFGF;③升高的IL1α和bFGF来源于被菌体活化的角朊细胞而非来源于菌体自身.以上结果有助于理解作为皮肤正常菌群的糠秕孢子菌为何可引起多种炎症性皮肤病.

Abstract: In order to study the interaction of Malassezia furfur to cultured human keratinocytes,we established a M.furfur keratinocyte co culture system by which the morphologic and functional investigations were combined closely.These were as follows:①The co cultured materials were observed regularly by scanning and transmission electron microscopy.②The effects of the conditioned media on the keratinocyte growth were monitored by a fluorophotometric assay with Alamar blue.③IL 1α and bFGF in the conditioned media were quantitated using ELISA kits.It was found that:①the organisms were adhered to and phagocytized by keratinocytes with a time dependent manner,and the keratinocytes revealed obvious morphologic changes.②the conditioned media could promote growth of keratinocytes,and higher levels of IL 1α and bFGF were detected in the media.③the increased IL 1α and bFGF were secreted from the organism activated keratinocytes rather than from the organisms themselves.The results are helpful to understand why the normal skin flora,M.furfur,could induce various inflammatory skin disorders.
