1,25-双羟维生素D3和MC903对培养人角朊细胞白介素1α及白介素8分泌的影响

Regulatory Effects of 1,25-dihydroxyvitamin D3 and its Analogue MC903 on the Secretion of Interleukin-1α and IL-8 by Human Keratinocytes in vitro

摘要: 1,25-双羟维生素D3[1,25(OH)2D3]及其衍生物钙泊三醇(MC903)对银屑病有治疗作用。我们用体外培养人角朊细胞研究了1,25(OH)2D3和MC903对角朊细胞白介素1α(IL1α)及白介素8(IL8)分泌的影响。1,25(OH)2D3和MC903本身对未受刺激的角朊细胞IL1α分泌无明显影响。用佛泊脂PMA和脂多糖(LPS)刺激角朊细胞,使得IL1α分泌显着下降和使IL8分泌显着升高。同时加入1,25(OH)2D3和MC903可抑制这些变化,并显示剂量相关性。用肿瘤坏死因子α刺激角朊细胞,可使IL8分泌显着升高和使IL1α轻度升高。1,25(OH)2D3和MC903也可抑制这些升高。在本实验中,PMA/LPS刺激角朊细胞引起的IL1α和IL8分泌变化与银屑病皮损非常相似,提示1,25(OH)2D3和MC903的这些作用可能与它们对银屑病的疗效有关。

Abstract: The regulatory effects of 1,25-dihydroxyvitamin D3 and its analogue MC903 (calcipotriol) on the secretion of interleukin-1α and IL-8 by human keratinocytes were studied. Cultured human keratinocytes spontaneously secreted low level-of IL-1α and IL-8. 1,25-dihydroxyvitamin D3 and MC903 showed no effect on IL-1α secretion while slightly enhanced IL-8 secretion on non-stimulated keratinocytes. Stimulation with tumor necrosis factor-α(TNFα)caused marked increase in IL-8 and slight increase in IL-1α secretion. 1,25-dihydroxyvitamin D3 and MC903 significantly inhibited IL-1α and IL-8 secretion induced by TNFα. Stimulation with phorbol 12-myristate 13-acetate(PMA) and lipopolysaccharide(LPS) induced a significant increase in IL-8 level and decrease in IL-1α, a pattern similar to those seen in psoriatic lesions. 1,25-dihydroxyvitamin D3 and MC903 inhibited the increase of IL-8 and the decrease of IL-1α. These results suggest that the regulatory effects of 1,25-dihydroxyvitamin D3 and MC903 on IL-1α and IL-8 secretion by keratinocytes may contribute to their efficacy in the treatment of psoriasis.
