聚合酶链式反应快速检测白念珠菌

Use of polymerase chain reaction for detection of candida aibicans

摘要: 我们应用聚合酶链式反应(PCR)技术休外扩增编码白念珠菌细胞色素P450L1A1的基因片段,在7小时内可以检测出500μ1中含有30个醉母菌的临床标本.对22份临床标本(其中5份明性,17份阳性)同时进行的PCR检测和真菌学鉴定显示:两者具有较满意的一致结果.这17份阳性标本包括4份血标本,5份尿标本,6份痰标本及2份分泌物标本.另5份为阴性血标本.PCR技术在白念珠菌感染乃至其它真菌感染的快速诊断方面显示了良好的应用前景.

Abstract: Candida albicans is an opportunitsic pathogenic fungus causing local and systemic disease, the mortality has been increasing dramatically. Early detection and management of candidiasis are crueaal, but conventional methods of diagnosis need long time and have low positivity. The authors used the polymerase chain reaction (PCR) to amplify a fragment of fungal DNA coding for the cytochroma P450 L1AI of C.albirans in vitro. The technique provides unambiguous evidence of C. albicans in 7 hours with, a detection as low as 30 organisms. 22 clinical specimens were callectsd from 15 patients, in each case the result of PCR was confirmed by mycological method. PCR based detection of pathogenic organisms may have broad application in rapid screening for the presence of organisms, and provides rapid and sensitivt methnd for diagnosis.
