SLE患者NK细胞活性及淋巴细胞内环核普酸含量的研究

NK Cell Activity and Intracellular Conteats of Cyclic Nucleotides in Lyuaphocytes from Patients with SLE

摘要: 研究了26例SLE患者外周血单个核细胞(PBMC)内环核井酸(cAMP,cGMP)水平和NK细胞活性;并在此基础上观察了给予氨茶碱,地塞米松刺激后PBMC内eAMP水平及NK细胞活性变化. 结果表明对照组PBMC受氨茶碱或地塞米松刺激后CAMP水平明显升高而SLE组PBMC对刺激仅有轻微反应.氨茶碱、地塞米松对正常人NK细胞活性有明显抑制作用,但在SLE患者中未观察到此现象.提示SLE患者的CAMP代谢过程存在某种固有损伤,从而导致SLE琳巴细胞内依赖cAMP途径的免疫调节机制受损,导致NK细胞活性低下.

Abstract: The NK cell activity and cyclic nucleotides levels (CAMP, cGMP) in lymphocytes of peri-pheral blood mononuclear cells (PBMC) of 20 controls and 26 patients were studied. The diagnosis of all the patients were consistent with ARA criteria. The cAMP levels between the controls and patients showed no significant difference before dexamethasone and amino phylline treatment. The lymphocytes from the patients elicited a ditninished response to dex-amethasone and aminophylline, but the lymp-hocytes from the controls showed a significant increase of CAMP level by this two agents. cGMP level of lymphocytes from the patients was signifieantly higher than that from the controls. Dexamethasone and aminophylline induced significant inhibition of NK cell activity in the controls, while this phenomenon was not observed in the patients. Though our study is preliminary, it may seggest that cynic nucleotides play a conspicuous role in the pathogenesis of SLE. There txray be an intMnsic impairment of the CAMP dependent regulatory mechanism in PSMC from the patients, which may result in abnormal NK cell activity.
