体外重建皮肤与正常人皮肤基本特征的对比研究

A comparative study between human skin equivalent and normal human skin

摘要: 目的 用正常人的皮肤角质形成细胞和黑素细胞在体外重建皮肤,为将来用于临床、生物及药理学研究奠定基础.方法 从正常儿童的包皮分离、培养角质形成细胞、黑素细胞和成纤维细胞.用成纤维细胞和I型鼠尾胶原制备真皮类似物.将角质形成细胞和黑素细胞接种到真皮类似物表面进行培养,制备表皮类似物.将重建的皮肤类似物和正常人皮肤进行对比研究.结果 皮肤类似物的组织形态包括基底层、基底上层、角质层与正常人皮肤相似.与基底膜形成有关的标志,在皮肤类似物和正常人皮肤中的表达是相似的.角蛋白10、泛角蛋白在皮肤类似物和正常人皮肤中的表达也是相似的.结论 组织形态和所检测的各种标志的表达显示,该皮肤类似物与正常人皮肤相似.

Abstract: Objective To develop a human skin equivalent model (SE) using normal keratinocytes (KCs) and menalocytes (MCs),which would serve as the basis for future clinical,biologic and pharmacologic studies.Methods KCs,MCs and fibroblasts (FBs) were cultured after neonatal foreskins were taken from routine circumcision.Cultured fibroblasts were mixed with rat tail type I collagen to form a dermal equivalent.Normal KCs and MCs were seeded on the dermal equivalent surface and grew to fuse to form an epidermal equivalent,which was cultured at the air liquid interface a week later to promote the differentiation and cornification of the epidermis.After being harvested,the SE and normal human skin were both assessed for overall morphology,presence of basement membrane components and expression of different markers.Results The histology of the basal layer,suprabasal layer and horny layer of SE was similar to that of the normal skin.When the markers related to the basement membrane zone were evaluated,similar results were observed.Pan-cytokeratin and keratin 10 both expressed similarly between the SE and normal skin.Conclusion The evaluation of the histology and the expression of different skin markers demonstrate that SE is similar to normal skin.
