淋球菌染色体mtrR基因突变对淋球菌多重耐药性的影响

Effect of mtrR Gene Mutation on Multiple Antibiotic Resistance of Neisseria Gonorrhoeae

摘要: 目的 对淋球菌的染色体耐药机理进行了一定的探索。方法 聚合酶链反应及Sanger双脱氧链终止法,测定淋球菌mtrR基因的碱基序列,并做相应淋球菌的药敏试验。结果 具有青霉素耐药性的淋球菌毫无例外都有mtrR基因的单碱基突变,而耐壮观霉素的淋球菌在mtrR基因上有两个位点突变。结论 淋球菌染色体上mtrR基因的突变会引起淋球菌多重耐药性的产生,增加淋球菌对脂溶性细胞毒因子如脂溶性抗生素、去污剂类脂肪酸的抗性。

Abstract: Objective To investigate the mechanism of multiple antibiotic resistance of Neisseria Gonorrhoeae chromosome.Methods The mtrR gene sequence of Neisseria Gonorrhoeae was examined by polymerase chain reaction and Sanger′s method, the drug susceptibility testing of relevant strains was done.Results Among 37 stains resistant to penicillin, 21 were resistant to tetracycline, erythromycin and Triton X-100 simultaneouely; one of them was resistant to spectinomycin as well; 5 strains were resistant to erythromycin only. All penicillin-resistant strains had a single base mutation in mtrR gene, and spectinomycin-resistant strain mutated in two position of mtrR gene.Conclusion The missense and null mutations in the mtrR gene of Neisseria Gonorrhoeae resulted in the emergence of multiple antibiotic resistance.
