人毛乳头细胞体外培养生长曲线观察

摘要: 目的 观察人正常头皮毛囊真皮成分的细胞体外培养的生长繁殖曲线及表皮细胞生长因子对它们的影响.方法 采用胶原酶消化法培养毛乳头细胞和真皮鞘细胞,在4种培养基中进行传代培养,并在基础培养基、常规培养基+表皮细胞生长因子中进行3H-TdR掺入,测定细胞代谢情况.结果 毛乳头细胞和真皮鞘细胞、成纤维细胞的生长呈现三个时相,停滞期、对数生长期和缓慢生长期,表皮细胞生长因子对3种细胞的生长具有显著的促进作用,尤其是对真皮鞘细胞的作用最为显著.结论 常规培养基+表皮细胞生长因子对3种间质细胞的促生长作用表明,在器官型培养中对毛囊上皮细胞和真皮成分的细胞也会有促生长作用,为毛囊器官型培养和人工皮肤研究采用此培养基提供了实验依据.

Abstract: Objective To observe the growth kinetics of dermal papilla cells and dermal sheath cells from human hair follicles in vitro and the effect of epidermal growth factor (EGF)on them. Methods Dermal papillae which were isolated from human hair follicles by collagenase D and dermal sheath cells were cultured in vitro. Dermal papilla cells and dermal sheath cells were stimulated by epidermal growth factor,hydrocortisone and insulin,or granulocyte-macrophage colony stimulating factor and counted in different time. The 3H-TdR incorporation of these cells was determined in basal medium and routine medium+ EGF. Results The growth of dermal papilla cells,dermal sheath cells and fibroblasts all showed three phases: stagnant phase in first four days,log growth phase in 5th to 11th day and slow growth phase. The growth of three types of cells,especially the dermal sheath cells,was markedly stimulated by EGF. Conclusions The growth of three types of mesenchymal cells is markedly stimulated by EGF in routine medium+ EGF,which provides the experimental evidence that this medium can be used for the organotypic culture of epithelial and dermal cells from hair follicles and skin.
