隆凸性皮肤纤维肉瘤中CD34和血小板源性生长因子受体β的表达

Expression of CD34 and Platelet Derived Growth Factor Receptor β and Pattern of DNA Ploidy in Dermatofibrosarcoma Protuberans

摘要: 目的 探讨隆凸性皮肤纤维肉瘤(DFSP)中CD34、血小板源性生长因子受体(PDGFR-β)的表达及基因倍体的意义。方法 使用免疫组化结合图象分析法检测39例DFSP,30例皮肤纤维瘤(DF)皮损中CD34、PDGFR-β的表达情况。采用流式细胞技术分析样本细胞核基因倍体,计算其DI值。结果 CD34、PDGFR-β蛋白在DFSP中表达明显高于在DF的表达,差异具有统汁学意义(P<0.05)。直线相关分析表明,CD34、PDGFR-β蛋白的表达之间无相关性(P>0.05)。39例DFSP中9例为异倍体(23%),其余为二倍体,异倍体均为近二倍体(DI=1.16±0.14);30例DF中无1例异倍体。相关性分析发现,异倍体组PDGFR-β蛋白的表达较二倍体组高(P<0.05)。结论 CD34、PDGFR-β与隆凸性皮肤纤维肉瘤的发生、发展密切相关。

Abstract: Objective To investigate expression of CD34 and platelet derived growth factor receptor β (PDGFR-β) and pattern of DNA ploidy in dermatofibrosarcoma protuberans (DFSP), and to study their significance and relationship. Methods The expression of CD34 and PDGFR-β was detected by immunohistochemical SP method in paraffin-embedded specimens from 39 DFSPs and 30 dermatofibromas (DFs). The findings were quantitatively analyzed by image system. The pattern of DNA ploidy was detected by flow cytometry. Results The expression of CD34 and PDGFR-β in the DFSPs was higher than that in the DFs (P<0.05). Nine of 39 cases of DFSP (23%) were heteroploidy (near diploidy, DI=1.16±0.14), while none of the 30 cases of DF was heteroploidy. The expression of PDGFR-β in heteroploidy DFSPs was higher than that in ploidy DFSPs (P<0.05). Conclusions CD34 and PDGFR-β are related with the occurrence and development of DFSP. Moreover, the expression of PDGFR-β is related with the malignancy of DFSP.
