催乳素对系统性红斑狼疮患者单一核细胞产生IgG和抗dsDNA抗体的影响

Human Prolactin Enhances the Production of IgG and Anti-dsDNA Antibody in Peripheral Blood Mononuclear Cells from Patients with Systemic Lupus Erythematosus

摘要: 目的 探讨催乳素(PRL)对系统性红斑狼疮(SLE)患者外周血单一核细胞(PBMC)产生抗体和自身抗体的影响。方法 体外培养SLE患者和正常人PBMC,用催乳素(PRL)和白介素2(IL-2)、白介素10(IL-10)和植物凝集素(PHA)刺激外周血淋巴细胞,采用3H-TdR掺入法测定淋巴细胞增殖情况,用ELISA测定PBMC分泌的IgG型抗体和抗dsDNA抗体。结果 ①10-9mol/LPRL可刺激SLE患者的PBMC增殖。②10-9mol/L的PRL分别可以刺激SLE患者PBMC分泌IgG型抗体和抗dsDNA抗体,且活动期患者显著高于静止期患者和正常人对照(P<0.01)。③PHA+PRL、IL-2+PRL刺激SLE患者PBMC分泌IgG型抗体和抗dsDNA抗体分别显著高于PRL组(P<0.01);抗IL-2抗体+PRL组低于PRL组,差异有显著性(P<0.01);IL-10+PRL组与PRL组间差异无显著性(P>0.05)。结论 PRL能够刺激体外培养的SLE患者PBMC分泌IgG型抗体和抗dsDNA抗体;PHA、IL-2与PRL有协同作用。

Abstract: Objective To study the effect of human prolactin (PRL) on total IgG and anti-dsDNA antibody production in peripheral blood mononuclear cells (PBMC) from patients with systemic lupus erythematosus (SLE). Methods PBMC from SLE patients and control subjects were cultured, with the stimulation of PRL, interleukin 2 (IL-2), interleukin 10 (IL-10) and phytohemagglutinin (PHA). 3H-thymidine (3H-TdR) incorporation assay was used to study the proliferation of PBMC. Total IgG and anti-dsDNA antibodies in the cultured supernatants were measured by enzyme-linked immunosorbent assay (ELISA) in 19 patients of SLE and in 6 control subjects. Results ①The proliferation of PBMC in vitro was enhanced by 10-9 mol/L PRL in 19 patients of SLE. ②The production of IgG and anti-dsDNA antibodies in PBMC from 10 active SLE patients was much higher than that of 9 inactive patients (P<0.01) and that of 6 control subjects (P<0.01). ③The production of IgG and anti-dsDNA antibodies in the costimulation of anti-interneukin 2 monoclonal antibodies (anti-IL-2 mAb) plus PRL was significantly lower than that stimulated of PRL in PBMC from 19 patients; on the other hand that costimulated by IL-2 plus PRL group or PHA plus PRL group significantly higher than that only by PRL. There was not significant between IL-10 plus PRL stimulation group and PRL stimulation group. Conclusion These results suggest that PRL may increase the production of IgG, including IgG type anti-dsDNA antibodies in the PBMCs of SLE patients; and IL-2 plus PHA can enhancesuch an effect.
