流式细胞计数用于念珠菌的快速药敏试验研究

摘要: 目的 探索应用流式细胞计数(FCM)进行念珠菌快速药敏试验.方法 参照M27-A标准化方案,应用FCM检测氟康唑(FCZ)对21株念珠菌的抗菌活性,药物作用时间3h,非渗透性荧光染料碘化丙啶(PI)染色0.5h.根据不同药物浓度作用下各药物浓度管荧光强度的梯度变化趋势,将平均荧光均道值大于生长对照管60%的最小药物浓度设定为最小抑菌浓度,即MIC值.并将结果与M27-A标准化方案进行比较.结果 FCM所测MIC值范围在0.125~64μg/mL之间,M27-A标准化方案所测MIC值在0.25~64μg/mL之间,统计学检验两者差异无显著性(P>0.05),结果有很好的一致性.FCM试验全过程仅需4h左右.结论 FCM药敏试验具有快速、敏感、可靠的优势.

Abstract: Objective To develop a rapid method for estimating the activity of antifungal drug by flow cytometry (FCM). Methods Referring to M27-A the minimum inhibitory concentration (MIC) of 21 Candida isolates to fluconazole (FCZ) was detected. The samples were incubated with FCZ in different concentrations for 3 hrs. Then they were stained by a membrane-impermeable fluorochrome, prepidium iodide (PI), for 0.5 h. According to the fluorescence intensity of different drug concentrations, MIC was defined as the lowest drug concentration in which the mean fluorescence intensity was 60% higher than that in the active growth control. Results The MIC detected by flow cytometry susceptibility testing (FCST) ranged from 0.125 to 64 μg/mL and the MIC of M27-A ranged from 0.25 to 0.64 μg/mL, there was no significant difference in MIC between FCST and M27-A (P>0.05). In this study MIC result could be detected by FCST within 4 hrs. Conclusion FCST is a rapid, accurate and sensitive methed for Candida MIC determination.
