皮肤恶性黑素瘤皮损STAT3表达及活化STAT3水平的研究

摘要:

目的 研究皮肤恶性黑素瘤(CMM)皮损中STAT3和活化STAT3(p-STAT3)的表达及其意义。方法 采用免疫组化聚合物酶标二抗法,检测22例CMM皮损和23例皮肤色素痣STAT3表达和p-STAT3水平,分析其对CMM淋巴结转移的影响。 结果 CMM皮损STAT3表达显著高于皮肤色素痣(阳性率分别为95.5%和56.5%,χ2 = 9.23,P < 0.05),且其p-STAT3表达水平亦显著高于皮肤色素痣(阳性率分别为90.9%和43.5%,χ2 = 11.38,P < 0.05)。CMM皮损STAT3表达增加对CMM淋巴结转移与否无明显影响(P > 0.05),而p-STAT3水平升高为预示CMM淋巴结转移的危险信号(OR = 1.88,95% CI为1.05 ~ 3.38,P < 0.05)。 结论 CMM细胞STAT3的表达增加和活化水平升高在CMM发病机制中可能起重要作用,并且其活化水平升高有助于CMM的侵袭和转移。

Methods The s of STAT3 and p-STAT3 were detected with an immunohistochemical method using polymer enzyme-labeled secondary antibodies in skin specimens from 22 CMM lesions and 23 pigmented nevus lesions. The effects of STAT3 and p-STAT3 s on lymph node metastasis of CMM were evaluated. Results The rate of STAT3 was significantly higher in CMM lesions than in pigmented nevus lesions (95.5% vs. 56.5%, χ2 = 9.23, P < 0.05), so was that of p-STAT3 (90.9% vs. 43.5%, χ2 =11.38, P < 0.05). STAT3 in CMM lesions had no obvious effects on lymph node metastasis of CMM (P > 0.05), while the elevated of p-STAT3 in CMM lesions was a predictive factor for lymph node metastasis in CMM (OR = 1.88, 95% CI: 1.05 - 3.38, P < 0.05). Conclusion Increased of STAT3 and p-STAT3 in CMM may play important roles in the pathogenesis of CMM, and elevated p-STAT3 may contribute to invasion and metastasis of CMM.
