强脉冲光对BALB/c小鼠皮肤胶原的影响

摘要: 目的 研究强脉冲光对BALB/c小鼠真皮内胶原含量以及前胶原基因表达水平的影响.方法 使用强脉冲光照射BALB/c小鼠背部皮肤,取各时间照射组和未照射组皮肤进行组织学观察,包括病理切片H-E染色和Ⅰ型、Ⅲ型胶原免疫组化染色,同时提取皮肤组织RNA,逆转录PCR检测前胶原基因的表达.结果 照光后第1周内皮肤胶原染色与未照射部位比较无明显差别,2周后照射部位真皮开始增厚,并进行性增多至第8周;免疫组化染色Ⅰ、Ⅲ型胶原明显增多(P<0.05).2周后小鼠皮肤Ⅰ型、Ⅲ型前胶原mRNA表达水平也较对照组明显上调(P<0.05),且表达水平随照光后时间的延长呈上升趋势.结论 强脉冲光照射小鼠皮肤后可刺激真皮胶原含量增加.

Abstract: Objective To investigate the effects of intense pulse light(IPL)on the content of collagens and mRNA expression of procollagen in BALB/c mouse skin.Methods The BALB/c mice dorsal skin was irradiated by IPL.Skin specimens were taken at different time points after irradiation.Histopathological changes were observed by hematoxylin-eosin-staining,quantitative assessment of collagenⅠand collagen Ⅲ was performed with immunohistochemical staining,and mRNA expression levels of procollagen were detected by RT-PCR.Results After the irradiation,no obvious change was observed for the staining intensity of collegen at 1 week;however,the thickening of dermis began at 2 week,and continued until 8 week.The staining of collagenⅠand Ⅲ was also stronger in IPL-irradiated regions than in sham-irradiated areas at 2 week(P<0.05).Compared to the sham-irradiated group,the mRNA expression levels of procollagenⅠand Ⅲ were increased from 2 week up to 8 week after IPL irradiation(P<0.05),and this up-regulatory effect of IPL was time-dependent.Conclusion IPL irradiation can stimulate collagen production in mouse dermis.
