白念珠菌种特异性DNA探针的制备与鉴定

摘要: 应用DNA克隆技术成功地克隆到经EcaRI消化的自念珠菌(白念)2.6kb片段并标记成探针,与数种念珠菌及人白细胞IAN人在NC膜上作斑点杂交发现:此探针只能与白念珠菌及类星型念珠菌杂交呈强阳性,与其它念珠菌及人白细胞DNA杂交均阴性,提示可用于白念珠菌病原体的检溯及菌种的鉴定,并为系统性念珠菌病基因诊断的研究打下基础.

Abstract: An attempt is made to produce DNA probe that only hybridizes with the DNA from Candida albicans.Total DNr1 was iaolated from the cells of Candida albicans and digested with EcoRI restriction enzyme.A fragmont(2.6 Kb) was cloned into the EcoRI site of the plasmid vector pBR322 and replicated in E.coli K802.The candidal fragment was labelled with digoxijenin-dUTP(DigdUTP) by random primed DNA labelling method and used to probe dot blots of DNA from several Candida spacious and human leukocytes.Only the DNA from Candida albicans and a strain of Candida steliatoidea was found to contain sedtrences of significant homology for hybridization.From tlue above results,it is concluded that this cloned fragment may passibly be used as a DNA probe for detecting and identifying the presence of Candida albicans.
